- 黑生101是利用花粉管通道技术育成的第一个大豆品种,利用132个随机引物对黑生101及其供体、受体的DN A进行RAPD扩增,其中3个单引物和2个双引物组合起来,均能在后代黑生101中扩增出与供体相同而与受体不同的DN A片段,证明黑生101中确有供体DN A片段的导入。
- Abstract:
- Heisheng 101 w as the fi rst soybea n cul tiva r obtained f rom po llen tube cha nnel. Weused RAPD to ampli fy the to tal DN A of Heisheng 101 and it s dono r and recipient by 132ra ndom primers. The DN A polymo rphi sm resul ts of RAPD molecula r verificatio n indicated: Ei ther 3 sing le random primer or 2 do uble primers combina tions can be amplifiedpolymo rphic DN A f ragment s f rom Heisheng 101. The results proved that Hei sheng 101w as t ruly obtained f rom the t ransforma tion prog eny of it s do no r DN A fragment s int roducedinto recipient.
