- 以大豆茎尖作为材料, 采用MSB5(MS无机+B5 有机)培养基附加不同浓度的6 -BA 诱导不定芽分化, 建立了以茎尖为外植体的大豆组培高效再生植株体系。不同品种研究结果表明:预培养时的6 -BA 浓度对不定芽分化的数量以及芽的伸长有直接影响, 低浓度的6 -BA 产生芽数量少, 变异系数小, 有利于不定芽的伸长, 高浓度的6 -BA 产生芽数量多, 变异系数也小, 不易伸长, 在3 .0mgL 浓度的6 -BA 处理比较理想, 变异系数较大, 有效芽多。不同品种对6 -BA 的耐受程度也存在一定差异, 合适的6 -BA 浓度有使差异减小的趋势。
- Abstract:
- Tip shoot of soybean (Glycin max L .)were selected as explant and precultured in MSB (inorganic of MS +organic of B5)medium with different concentration of 6 -BA to inducing buds .The high frequency regeneration system oftissue culture of soybean was established .The result indicated that concentration of 6 -BA of preculture had a direct influenceto the number of buds and elongating of buds .Low concentration of 6 -BA produced a few buds , and the coeffecientof variability was small , but benefit to buds’ elongation ;high concentration of 6 -BA producdmany buds , the coeffecientof variability was small too , but not easy elongation .The treatment with 3.0mg L of 6 -BA was better , the coeffecientof variability was large , effective buds were more .Ditterent soybean varietes had difterent responses to the concentrationof 6 -BA , high concentration of 6 -BA tended to reduce the difference .
