- 将大豆内源参照基因( Lectin) 、外源基因P-CaMV 35S、CP4-EPSPS 和T-NOS 作为多重PCR 检测参数,建立了一种多重PCR 检测抗草甘膦大豆GTS40-3-2 的方法。反应条件优化结果表明: 多重PCR 的适宜退火温度为58℃,适宜引物终浓度( μmol·L - 1 ) 配比为: 0. 1∶0. 2∶0. 2∶ 0. 2。采用已知样品对多重PCR 体系进行验证,检测结果可靠,证明该多重PCR 体系为一种有效的抗草甘膦大豆GTS40-3-2 的检测方法。
- Abstract:
- In order to establish a multiplex PCR ( polymerase chain reaction) method detection for Roundup-Ready soybeanGTS40-3-2,the soybean endogenous reference gene ( Lectin) ,exogenous gene CaMV 35S promoter,CP4-EPSPS and NOSterminator were used as detection parameters. The optimization results showed that the optimum annealing temperature of multiplePCR was 58℃,and the optimum final concentration of primers( μmol·L - 1 ) ratio is 0. 1∶ 0. 2∶ 0. 2∶ 0. 2. The multiplexPCR method is confirmed to be reliable through known samples. And it is proved that the multiplex PCR system is an effectivemethod for detection Roundup-Ready soybean GTS40-3-2.
