向大豆导入几丁质酶基因的初步研究

- 以根癌农杆菌的Ti质粒为介导,将抗真菌病的几丁质酶基因,导入黑龙江省栽培大豆- 东农37号,吉林28号等14个品种。从子叶节和下胚轴诱导出丛生芽,并再生植株,经PCR和DN A分子杂交检测,确认为转化植株。用花粉管通道导入法,直接导入几丁质酶基因,共获223粒种子,经PCR和DN A分子杂交检测的113株大豆幼苗中,有7株呈阳性反应,证明几丁质酶基因已导入并整合到大豆的基因组中。
- Abstract:
- The chi tinase gene w as transferred into cultiva rs Do ng no ng 37, Ji lin 28, et al. w ithTi plasmid. Adventi tious nuds and reg ennera ted plant were inducted f rom co tylendonusnode a nd h ypoco tyl. PCR and DN A do t blo t ting prov ed tha t they w ere t ransferredplant. Puri fied plasmid PBin 437 with chitinase gene was also t ransferred into sev eralsoybeans va ridties by the pol len tube technique and 223 seeds were obtained. Among 113plants g ermina ted f rom these seeds 7 show ed positiv e reaction in PCR and DN A dotblo tting.
