RIL群体中大豆籽粒脂肪酸组分QTL分析

- 大豆是世界上主要的油料作物,其脂肪酸各组分的比例决定了大豆油的质量。本研究以“徐豆16 × 冀HJ117”构建的包含183个家系的重组自交系(Recombinant Inbred Lines,RILs)群体XH1617为材料,通过气相色谱法检测棕榈酸、硬脂酸、油酸、亚油酸、亚麻酸5种脂肪酸含量。表型变异分析显示,RIL群体的5种脂肪酸含量均呈近似正态分布,说明大豆5种脂肪酸含量均为典型的数量性状。通过QTL定位,共鉴定到13个与大豆脂肪酸含量相关的QTL位点,分别位于05、06、08、13、14、15和19号染色体上,LOD值为2.50~12.35,可解释的表型变异范围为6.1%~26.7%。其中6个QTL是新鉴定到的与大豆脂肪酸含量相关的遗传位点。另外,在15号染色体上鉴定到1个主效且协同调控大豆棕榈酸含量和油酸含量的遗传位点,Locus_15_POA,在标记Gm15_3702534_C_T~Gm15_4491175_T_C之间,LOD值为4.71~5.11,可解释的表型变异为11.20%~12.10%。相关性分析发现,除硬脂酸含量与油酸含量无显著性相关外,其他脂肪酸含量之间均具有显著相关性(P<0.05)。研究结果可为培育高油酸且低棕榈酸的高品质大豆提供遗传材料和理论依据。
