- 选用不同浓度的染料木素( GEN) 及二十二碳六烯酸( DHA) 作用于MCF-7 细胞,研究大豆中抗癌成分染料木素与不同比例的多不饱和脂肪酸( PUFAs) 联用,对乳腺癌细胞MCF-7 增殖及脂代谢基因表达情况的影响。根据DHA的有效浓度设定亚油酸( LA) 及LA∶ DHA( 10∶1) 混合PUFAs 作用浓度。将GEN 分别与DHA、LA 及LA∶ DHA( 10∶ 1)联合作用细胞,检测细胞生存率,并通过qPCR 方法检测细胞内过氧化物酶体增殖物激活受体γ( PPARγ) 、5 - 脂氧合酶( 5-LOX) 、环氧合酶- 2( COX-2) mRNA 表达情况。结果表明: 高浓度GEN( 300, 200, 100 μmol·L - 1 ) 及DHA( 400,300, 200 μmol·L - 1 ) 可抑制MCF-7 细胞的增殖,且随着时间增加抑制作用增强。而低浓度GEN( 50, 25 μmol·L - 1 ) 及LA 组及10∶1 组对细胞增殖有轻度促进作用。添加GEN 的G + L 组和G + 10∶1 组细胞的生存率显著降低,同时GEN可降低G + L 组细胞5-LOX mRNA 的表达量,且GEN 与LA∶ DHA( 10∶1) 联用,增加了细胞内PPARγ mRNA 的表达水平并抑制了COX-2 mRNA 的表达。试验证实GEN 可抑制高比例ω-6 PUFAs 对乳腺癌细胞增殖的促进作用,其作用机制主要是通过增加PPARγ mRNA 表达,抑制COX-2 mRNA 表达水平实现的。大豆的抗乳腺癌作用可能是由于GEN与PUFAs 在抑制乳腺癌细胞增殖方面产生了协同效应。
- Abstract:
- The effect of gentistein which is the anticancer component in soy,combined different ratios polyunsaturated fattyacids( PUFAs) on proliferation and lipid metabolic gene expression of MCF-7 cells was studied. MCF-7 was cultured with differentconcentration gentistein( GEN) and docosahexaenoic acid( DHA) . According to the effect concentration of DHA,we setlinoleic acid( LA) and mixed PUFAs’ concentration,which was 10∶ 1 LA to DHA. Then we combined GEN with DHA,LAand LA∶ DHA ( 10∶1) respectively,texted the survival rate and detect mRNA expression of peroxisome proliferators-activatedreceptor γ( PPARγ) ,5-lipoxygenase ( 5-LOX) and cyclooxygenase-2 ( COX-2) with qPCR. The results showed that highconcentration of GEN and DHA could inhibit proliferation of MCF-7 cells and the effect enhanced with time. However,lowconcentration of GEN could promote the cells proliferation mildly. LA and 10∶ 1 PUFAs promote the cells proliferation,butcell’s proliferation was inhibited when we added GEN. GEN could reduce 5-LOX mRNA level of MCF-7 cell which was underthe effect of LA. Meanwhile it inhibits proliferation of MCF-7 cell through promoting PPARγ mRNA level and inhibiting COX-2 mRNA level when the cell is cultured with GEN combined 10∶1 LA∶ DHA. GEN can inhibit the promoting effect of high ratioω-6 PUFAs’ proliferation of breast cancer cells. The mechanism is mainly through improving the expression of PPARγ mRNAand inhibiting COX-2 mRNA. The synergistic effect of GEN and PUFAs on inhibiting the proliferation of breast cancer cells,may be the reason for the anti breast cancer of soy.
