- 以周豆12为材料,研究了不同浓度的抗坏血酸(ASA)对盐胁迫下大豆愈伤组织生长的影响,以期为大豆抗盐胁迫研究提供参考。结果表明:盐胁迫下,大豆愈伤组织的相对含水量、相对干重增长速率、可溶性蛋白含量显著降低,Pro、MDA含量以及SOD、POD活性显著增加(P<0.01);施用ASA显著提高了盐胁迫下愈伤组织的相对含水量、相对干重增长速率、可溶性蛋白和Pro含量,SOD和POD活性也显著升高,膜脂过氧化产物MDA含量显著降低(P<0.01)。由此可见,ASA可以通过提高大豆愈伤组织的相对含水量、可溶性蛋白和Pro含量以及SOD、POD活性来维持细胞膜的稳定性,降低膜脂过氧化伤害程度,从而缓解盐胁迫对愈伤组织生长的抑制,并以8 mmol·L-1外源ASA缓解效应最好。
- Abstract:
- The response of soybean callus growth to ascorbic acid (ASA) under salt stress was studied, using soybean Zhongdou 12 as material. Under salt stress, the relative water content(RWC), soluble proteins content(SPC) and increase rate of dry weight(IRDW)were significantly decreased, the contents of proline(Pro) and malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and peroxidase(POD) were significantly increased(P<0.01). When applied ASA, the RWC, SPC, IRDW, Pro, as well as SOD and POD activities were all increased, while the MDA content was significantly decreased (P<0.01). The results showed that ASA could effectively keep cytomembrane stabilization and reduce the damage from membrane lipid peroxidation, by which could mitigate the harmful effects from salt stress and promote the growth of callus, and the optimal ASA concentration was 8.0 mmol·L-1.
