- 为有效控制大豆异黄酮缓释微丸胶囊的质量,建立其中一种有效成分金雀异黄素含量的测定方法。采用RP-HPLC法测定大豆异黄酮缓释微丸胶囊中金雀异黄素的含量,固定柱:ZOBAX SB-C18(5 μm,250 mm×4.6 mm);流动相:乙腈-0.1%H3PO4水溶液(40∶60);检测波长:262 nm;柱温:35℃;流速:1.0 mLmin-1;进样量5 μL,应用外标一点法以峰面积定量。金雀异黄素在20.88~208.8 ng之间线性关系良好,回归方程为Y=4.38X-2.69,R=0.999(n=6),平均回收率为99.97%,RSD为0.87%(n=9)。此法操作简便,结果准确可靠,精密度好,可用于大豆异黄酮缓释微丸胶囊中金雀异黄素的含量测定。
- Abstract:
- In order to validly control the quality of soybean isoflavone sustained-release pellet capsules,a method for determination of genistein in it was established using the method of RP-HPLC for determination.Chromatographic conditions include:column:ZOBAX SB-C18(5 μm,250 mm×4.6 mm);mobile phase:acetonitrile -0.1% H3PO4 solution (40∶60);detection:UV 262 nm;temperature:35℃;flow rate:1.0 mL min-1;sample size:5 μL.Using one point external standard method to quantitate.Genistein had a good linear relationship in the range of 20.88-208.2 ng,regression equation was Y=4.38X-2.69,R=0.999 (n=6).The average recovery of genistein was 99.97%,RSD=0.87%(n=9).The method is rapid,accurate,reliable and can be used to determine genistein in the soybean isoflavone sustained-release pellet capsules.
