大豆花叶病毒 5 个分离物的鉴定及外壳蛋白序列分析*

- 通过生物学纯化与血清学鉴定(ELISA)得到 5 个大豆花叶病毒(SMV)分离物, 利用 RT -PCR 法扩增其外壳蛋白(CP)基因片段并进行测序 ,结果表明 :5 个分离物 CP 基因全长均为 795 个核苷酸, 编码产生 265 个氨基酸 。同源性分析表明, 5 个分离物核苷酸序列同源性为 91. 1%~97. 1 %,由此推导的氨基酸序列同源性为 98. 1%~ 99. 2%。结合 5 个分离物在 8 个大豆品种上的致病性反应得出 ,5 个分离物在 8 个鉴别寄主上的致病性反应和序列差异的比较表明, 分离物在 8个鉴别寄主上致病性反应相差越大, 其 CP 基因氨基酸序列差异就越大, 由此证实 SMV 的 CP 基因与致病性反应之间有一定的联系 。
- Abstract:
- 5 SMV isolates were obtained by using biological purification and serology (DAS-ELISA). Thecoat protein gene regions were amplified by reverse transcription polymerase chain reaction (RT-PCT). Se-quences analysis showed that CP gene of the each SMV isolates had 795 nucleotides in length , encoding265 amino acids. Comparison of the sequences of CP genes between different isolates showed they shared91. 1%~ 97. 1%nucleotide acids identities and 98. 1%~ 99. 2 %amino acidsidentities with each other. Theresults also revealed that there was significant correlation between CP genes of SMV and symptom reactionof SMV on differential Hosts
