- 利用BSA 法对11个黑色种皮的大豆品种和 12 个黄色种皮的大豆材料的基因组 DNA 进行了 RAPD 分析, 供试的 100 个随机引物有 66 个产生了 RAPD 扩增产物 ,其中产生 RAPD 多态性的随机引物有 15 个 。在所产生的扩增产物中 , 获得了一个与大豆种皮色相关的特异 DNA 片段S79500 。同时,采用该标记对辽豆 10 号×PI437654 杂交后代种皮颜色有分化的群体和黄 、青 、黑、褐色种皮的大豆进行检测 ,经验证这个 RAPD 标记具有较高的重复性和稳定性 ,可用于辅助选育优良的黄色种皮的大豆新品种 。
- Abstract:
- BSA method was employed to analyze genome DNA of 11 black coat seed soybean and 12 yellowcoat seed soybean by RAPD method. The bands were produced for 66 primers among 100 primers. Therewere polymorphism bands for 15 primers. In all products amplified, a special DNA fragment , S79500 wasclosely related with the yellow coat of soybeans. The primer S79500 became a RAPD marker for the yellowcoat of soybean. In another hand, the S79500 also was used to test the filial generation with yellow ,green, black and brown coat from Liaodou 10 ×PI437654. The results showed that the S79500 was verystable and repeatable. The RAPD marker S79500 was useful to assistant the selection for soybean hybridbreeding.
