大豆根际高效自生固氮菌分离鉴定与特性研究

- 采用无氮培养基,从大豆根际土壤中分离获得4株具有固氮能力的菌株:LS01、LS04、LS05和LS06。根据菌落的形态特征及16S rDNA序列比对分析,结果表明:LS01属于Microbacterium sp,LS04属于Streptomyces sp,LS05属于Micromonospora sp,LS06属于Sarocladium sp,在无氮液体培养基中培养64 h生物量均达到最大且能够分泌吲哚乙酸(IAA)。利用乙炔还原法对4种菌的固氮酶活性进行测定,固氮酶活测定结果表明4种菌均具有较高的固氮酶活性。因此,Microbacterium sp LS01,Streptomyces sp LS04,Micromonospora sp LS05和Sarocladium sp LS06具有一定的固氮能力并能分泌IAA,在制备大豆生物菌肥方面具有较好的应用前景
- Abstract:
- Agricultural production in China is facing the challenges in high yield with high quality and environment protection. Authigenic azotobacter can promote plant growth, increase crop yield and quality, and have no contamination to environment, therefore, it will have broad applications in agricultural production. At present, it is a hot project to screen plant growth promoting rhizobacteria from specific crops as biological fertilizers.In the study, four strains were isolated and identified from plant rhizosphere, and named as LS01, LS04, LS05 and LS06, respectively.The analysis of enzyme activity found that all of them had capability on nitrogen fixation.The characteristics of morphology and 16S rDNA sequence analysis revealed that strain LS01 was classified as Microbacterium sp, strain LS04 was classified as Streptomyces sp, strain LS05 was classified as Micromonospora sp, and strain LS06 was classified as Sarocladium sp. The characteristics of their growth indicated that the maximum biomass were reached in 64 hours in no nitrogen medium. The further analysis showed that the four authigenic azotobacter had higher nitrogenase activity by use of acetylene reduction method.In the meantime, IAA quantitative analysis showed that they could secrete indole acetic acid. The data obtained in our study suggest that the four strains will have promising applications in producing bio-bacterial fertilizers and in promoting plant growth.
