- ?为研究大豆异黄酮对D-氨基半乳糖(GalN)和内毒素(LPS)联合所致小鼠急性肝损伤的保护作用,将试验小鼠按体重随机分为5组,即正常组、模型组、水飞蓟素组(阳性对照组)及大豆异黄酮高、低剂量组。每日给药1次,连续7 d。除正常组外,其余小鼠腹腔注射GalN及LPS建立急性肝损伤模型,苏木素伊红(HE)染色法观察肝组织病理学变化,ELISA法检测血清肿瘤坏死因子-α(TNF-α),比色法检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)以及肝脏一氧化氮(NO)、过氧化脂质(LOOH)、丙二醛(MDA)含量和Caspase-3、Caspase-8活化水平。结果表明:大豆异黄酮显著降低GalN及LPS联合诱发的急性肝损伤小鼠血清ALT、AST活性,减轻肝组织病理损伤,降低血清TNF-α和肝NO水平,降低肝LOOH和MDA水平,降低肝组织Caspase-3和Caspase-8活化水平。说明大豆异黄酮对GalN和LPS联合诱导的小鼠急性肝损伤具有保护作用,其机制可能与其抗炎、抗氧化活性和抗肝细胞凋亡作用有关。
- Abstract:
- ?For the purpose of studying the protective effect of soy isoflavones on acute liver injury induced by D-galactosamine(GalN)and lipopolysaccharide(LPS)in mice,the tested mice were randomly assigned to 5 groups:normal control,model control,silymarin(positive control),as well as high and low dose groups of soy isoflavones.Animals were treated once daily for 7 days.GalN and LPS were given intraperitoneally to the mice of groups,then the pathological changes was detected with hematoxylin & eosin(HE)staining,tumor necrosis factor-α(TNF-α)was detected by ELISA method,and the alanine aminotransferase(ALT),aspartate aminotransferase(AST),lipid hydroperoxide(LOOH),malondialdehyde(MDA),nitric oxide(NO),and the activation of Caspase-3 and Caspase-8 were detected by the colorimetric method.The results showed that the soy isoflavones reduced the serum ALT and AST,alleviated the hepatic lesions induced by GalN and LPS,decreased serum TNF-α and hepatic NO levels,reduced the hepatic LOOH and MDA contents,and decreased the Caspase-3 and Caspase-8 activations of liver tissues.It is suggested that soy isoflavones have protective effect on acute liver injury induced by GalN and LPS in mice,probably via the anti-inflammatory,antioxidative and anti-apoptotic ability.
