- 以大豆为原料,甲醇为浸提溶剂,5种大豆异黄酮提取率为指标,利用超声波辅助提取,通过单因素实验初步确定提取工艺条件。然后在单因素实验的基础上采用3因素3水平的响应面分析法,建立了总大豆异黄酮含量与各影响因子的回归方程,并得到以总大豆异黄酮提取率为响应值的响应面图和等高线图,进而得出最佳提取工艺条件为固液比1∶50、甲醇提取液浓度85%、提取时间45 min。在此条件下提取2次,大豆异黄酮的总提取率可达0.308%。
- Abstract:
- The objective of this study was to optimize the extraction conditions of soybean isoflavone,taking soybean as raw material,using the methanol as extraction solvent and ultrasonic assisted extraction method.The extraction parameters was preliminary determined through the single factor experiment.Then the 3 factors 3 levels of response surface analysis method was established using Design-Expert 7 experimental design software,and got the response surface chart and contour map with the response value of the total soybean isoflavones extraction efficiency.The result showed that the best extraction technology conditions was at the ratio of material and solvent 1∶50,with 85% methanol extract concentration and extracting for 45 min.On this condition extracting for 2 times,total soybean isoflavone extraction ratio could reach 0.308%.
