大豆异黄酮纯化工艺研究

- ?以大豆异黄酮纯度为考察指标,将醇沉结合乙酸乙酯萃取法与其他纯化方法进行对比,并进一步对醇沉法中工艺参数通过单因素试验及正交试验进行优化。结果表明:醇沉最佳工艺为浓缩液质量分数25%,乙醇浓度80%,搅拌时间13 min,静置时间5 h;经等体积乙酸乙酯萃取3次,异黄酮基本萃取完全。醇沉结合乙酸乙酯萃取法可得到含量高达16.83%的大豆异黄酮产品,比单纯醇沉法提高约4倍。
- Abstract:
- ?With the purity of soybean isoflavone as investigation index,the isoflavone purification method of alcohol precipitation combined with ethyl acetate extraction was compared with other methods.And then the alcohol precipitation parameters for the purification of isoflavones were optimized by single-factor and orthogonal experiment.The optimized conditions of ethanol precipitation were,mixing 25% mass fraction of isoflavone extract with 80% ethanol for 13 min,and then standing for 5 h;After 3 times extraction with ethyl acetate,isoflavones could be extracted completely.Under the new technology of alcohol precipitation and ethyl acetate extraction,the purification of isoflavones in final product was 16.83%,increased nearly 4 times than separate ethanol precipitation.
