- 菜豆荚斑驳病毒是对大豆经济具有重要影响的病毒 。根据已报道的菜豆荚斑驳病毒外壳蛋白基因序列 ,设计出一对特异性引物 ,采用 RT - PCR法 ,可有效地扩增出 BPMV 不同亚组的 4 个分离物,得到 650 bp 大小的 PCR产物。这种方法能成功地检测出大豆病叶中的 BPMV ,结果和酶联检测是一致的 ,而对大豆种皮中 BPMV 的检测效果不佳 ,建议采用酶联的方法进行检测。
- Abstract:
- Bean pod mottle virus is an economically important virus that infect soybean. A pair of specificprimers was designed based on the conserved region of the coat protein gene of bean pod mottle virus. To-tal RNA was extracted from infected soybean leaves and was reverse transcribed to cDNA. The cDNA wassubjected to PCR using the forward and reverse primers. PCR products of 650 bp were generated from allisolates tested. The RT - PCR method could be used to detect BPMV - infected soybean leaves with satis-fied results that were confirmed by DAS - ELISA assay. We recommended ELISA assay to detect BPMV -infected soybean seed rather than RT - PCR .
