大豆异黄酮提取条件和大豆蛋白质分离工艺研究*

- 以豆粕为原料 ,以总大豆异黄酮为目标产物 ,用正交试验设计研究了用乙醇提取大豆异黄酮的较优条件 ,70 %(V /V)乙醇 ,固液比(豆粕 ∶溶剂)1 ∶10(W /V),70 ℃下提取 3 次, 每次回流时间1. 5h 。在室温下, 采用盐析和等电点沉淀工艺分离除去提取液中的大豆蛋白质。试验表明, pH 值在 4. 1±0. 1 下 ,加入 3. 0%(m /v)的 ZnCl 2 ,大豆蛋白质的分离除去率最高。
- Abstract:
- The conditions of extracting soybean isoflavones from soybean meal with ethanol was proposedand investigated in this paper. On the purpose to get the yield of total soybean isoflavones as high as possi-ble , with orthogonal experimental methodology , the optimum parameters of extracting isoflavones wereobtained , i. e. 70 %(V /V)ethanol , soybean meal weight to solvent volume at 1 ∶10 , temperature 70℃,time 1. 5h and 3 times repeat. The protein was separated from the pre - extracting liquid with salting - outand isoelectric precipitation at room temperature. The results indicated that the separation quotiety of pro-tein was the highest with the ZnCl 2 (3. 0%,m /v)at pH 4. 1±0. 1.
