- 利用Solt-Faber法制备大鼠肝癌初期模型,同时以100 mg·kg-1大豆异黄酮和皂甙干预42 d。然后处死动物,制备肝匀浆及肝线粒体提取液。以比色法测定丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、γ-谷酰胺转肽酶(γ-GT)、谷胱甘肽-S-转移酶(GST)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)含量。结果表明,大豆异黄酮和皂甙能降低肝癌发生初期大鼠肝脏ALT、AST、γ-GT和GST活性,升高肝脏及肝线粒体SOD、CAT、GSH-PX活性和降低MDA水平。提示,大豆异黄酮和皂甙具有减轻肝癌发生初期大鼠肝细胞损伤,降低其氧化应激的作用。此作用有助于预防肝癌的发生。
- Abstract:
- The early stage of hepatocarcinogenesis was established by the method of Solt and Farber,and the rats were administered with 100 mg·kg-1 of soybean isoflavones and saponins for 42 days.At the end of experiment,the animals were killed and the liver homogenates and liver mitochondria were extracted.Then the activities of alanine trasaminase(ALT),aspartate aminotransferdse(AST),γ-glutamyl transpeptidase(γ-GT),glutathione S-transferase(GST),superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX),as well as the content of malondialdehyde(MDA)were detected by colorimetric method.The results showed that the administration with soybean isoflavones and saponins reduced the hepatic activities of ALT,AST,γ-GT and GST,increased the SOD,CAT and GSH-PX and decreased the MDA of liver and liver mitochondria of rats in early stage of hepatocarcinogenesis.It is suggested that soybean isoflavones and saponins could elevate hepatic injury and reduce hepatic oxidative stress of rats in early stage of hepatocarcinogenesis.This function will be helpful for the prevention of liver cancer.
