Abstract:
To establish methods for the determination of Daidzein and Genistein in Semen Sojae Praeparatum by high performance liquid chromatography and total isoflavones by ultraviolet spectrophotometry.The established HPLC conditions were:an Agilent HC-C18 column(250×4.6 mm,5.0 μm)was applied;the mobile phase was MeOH∶0.1%HAc=50∶50;the temperature of column was 30℃;the flow rate was 1.0 mL·min-1;detective wavelength was set at 261 nm. Wavelength at 261 nm of ultraviolet spectrophotometry was adopted to determine the absorbance of samples solution.The results showed that the retention time of Daidzein and Genistein was 9.79 and 14.87 min,with the average recovery 98.90% and 100.53%,respectively.The integral value of peak area has linear relationships with sample amount of Daidzein or Genistein(r>0.9999).Total isoflavones from 1.44 to 7.20 μg·mL-1 showed a linear relationship and the average recovery was 102.26%.The methods are simple and easy to evaluate the quality of Semen Sojae Praeparatum.
