Abstract:
To study the effect of Semen Sojae Praeparatum(SSP)extract on the NO/NOS system in T2DM rats.A T2DM rat model was developed by means of high-sugar,high-fat feed and raperitoneal injection of low-dose streptozotocin;after 56 days drug administration,the levels of fasting blood-glucose(FBG)and fasting serum insulin(FIN)in the rats were detected,insulin sensitivity index(ISI)was calculated;serum tumor necrosis factor-α(TNF-α)and nitric oxide(NO)level were detected by radioimmunoassay.And the expression of aortic endothelial nitric oxide synthase(eNOS)and inducible nitric oxide synthase(iNOS)mRNA was detected by RT-PCR.The levels of FBG,FIN,TNF-α and NO in T2DM rats were decreased lowered substantially,ISI was increased significantly,iNOS mRNA expression was markedly reduced,and eNOS mRNA expression was not obviously affected by SSP extract.These results suggest that SSP extract can affect the expression of iNOS mRNA and the level of NO in the T2DM rats,and may play a role in the protection against macrovascular disease of large vessels in T2DM rats.
