Abstract:
To investigate the effects of Semen Sojae Preparatum(SSP)on apoptosis,and gene expression of p53 and Fas in aortic smooth muscle cells(ASMCs)in early Atherosclerosis(AS)rats,and to explore the possible mechanism.Rat models with early AS were induced by feeding fatty foods,meanwhile rats in two treated groups were orally administered with extract of SSP for 10 weeks.All rats were killed and samples of ASMCs harvested.The cellular morphology was observed by light microscope and ultrastructure by transmission electron microscopy;apoptosis and gene expression of p53 and Fas were detected by flow cytometry.Compared with normal group,the apoptosis rate and the proliferation index(PI)in model group were obviously increased(P<0.01),with higher gene expression of p53;moreover compared with model group,the apoptosis rates in two treated groups(20 gkg-1,40 gkg-1)were higher(P<0.01),the proliferation index was lower(P<0.01),and the gene expression of Fas was increased,but p53 was decreased(P<0.05);histologically,the aortic damage was remarkably improved with SSP.These results suggest that SSP can adjust the balance between apoptosis and proliferation of ASMC,which may relate to gene expression regulation of p53 and Fas.
