Abstract:
Plant transformation is an important approach in genetic engineering breeding,great advances have been achieved in soybean transformation recently. In order to broaden soybean receptor genotypes and enhance the application value of transgenic soybean,new soybean lines Hajiao 5337 and Hajiao 5489 were used to optimize the regeneration of the cotyledon node organ. Different concentration of 6-BA and IBA were used in the shoot induction medium,the ratio of shooting was analyzed using the orthogonal analysis. The cotyledon node grown in the germinated medium with 6-BA(named G1)and the germinated medium without 6-BA(named G2)were used as controls,respectively. It indicated that G1S7 and G2S4 was the optimum combination for the cotyledons of Hajiao 5337 in germinating medium and shooting induction medium,while G1S4 and G2S4 for Hajiao 5489. The adoption of the delay screening,the optimal screening concentration of Glufosinate-ammonium to Hajiao 5337 and Hajiao 5489 was 3.5 mgL-1in shooting induction medium.
