Abstract:
lkt50 gene was introduced into soybean cultivar Dongnong 48 and strain Dongnong 96-02 via pollen tube pathway.The transformed plants were primary screened from seedling by spraying kanamycin,and further the transformed plants was determined by molecular biological methods including PCR,Southern blot and RT-PCR.The results indicated that the identification of 14 Km resistant plants was determined by PCR with the use of the primers of nptII and lkt50,and seven and two plants were found to be positive respectively.Furthermore,the detections by Southern blot and RT-PCR revealed that the two plants were found to be positive.The pollen tube pathway is an effective,feasible method for soybean transgene.
