Abstract:
The 14 w ild soybeans and cultivated soybeans w ere studied in this experiment .Hypocotyls w ere usedas explants .Callus w as induced at MS +2 , 4 -D2 .0mg/L +6 -BA0 .5mg/L + sucrose 3 % + agar 0 .8 %.The salt tolerance of callus w as observed on different medium containing 0(as contrast), 0 .1 %, 0 .2 %, 0 .3 %,0 .4 %, 0 .5 %, 0 .6 %, 0 .7 %, 0 .8 %NaCl .The experimental materials were planted also in soil pond containing0 .0 % 、0 .3 %NaCl.The results showed that salt tolerance of callus from Zhonghuang 4 , Tongnong 5 and Jilin35w as st ronger .Among different materials , the difference of salt tolerance w as remarkable .The higher salt tolerancematerials could grow well o n medium containing 0 .8 %NaCl , the lower salt tolerance materials could onlyg row on medium containing 0 .4 %NaCl.Sui table concent ration for identifing salt tolerance of soybean callus was0.6 %NaCl in this experiment.MDA content of plant s in 0 .3 %NaCl w as higher than that in control.MDAcontent of plant s grow n on 0 .4 %NaCl soil pond at different time appeared dif ferent varies wi th the increase ofsalt st ress day s.The co rrelation between MDA content and callus salt tolerance w as not found in this paper .
