Abstract:
The 14 genoty pes of wild soy bean growing at different condi tions in eastern of Hebei province w erestudied in this paper .Hypocotyls and co tyledon were used as ex plants , through tissue culture , calli w as inducedon 4 kinds of medium containing different composition .The calli were subcultured on D4 medium for 4 times.The salt -tolerance of calli was observed on dif ferent medium containing 0 .0 , 0 .2 , 0 .4 , 0 .6 , 0 .8 , 1 .0 %NaCl.Abili ty of salt -tolerance w as identif ied through g row th amount , quality , becoming brow n and death ratio of calli.The result s show ed that MS +2 , 4 -D2 .0mg/L +6 -BA0 .5mg/L +sucrose 5 %+ag ar 0 .8 % is the bestmedium for callus induction .Salt -tolerance ability of calli had obvious dif ferent among different w ild soybean .The stronger salt -tolerance w ild soybean could survive at the concent ration of 0 .8 %NaCl .The ability of salt -tolerance of 6 、7 、10 、11 、12 、14 w as stronger , the g row th amount and quality of calli descended with the rising ofconcent ration of NaCl.The experiment indicated that screening concent ration fo r salt -tolerance of w ild soybeanis 0 .6 %NaCl.Generally , the w ild soybeans f rom saline soil have st ronger abili ty of salt -tolerance ,w here as,the w ild soybean f rom non -saline environment have very strong existing ability in NaCl medium , too .There w as no t complete co rrespondence relatio n betw een g row th environment and salt -tolerance ability at cell level.The results sug gested that the relation of salt -tolerance ability at cell level and plant level should study further .
