Abstract:
To establish a high frequency regeneration system for the gene t ransfo rmation of soybean(Glycinemax (L .)Merr .), cotyledonary nodes f rom ten varieties w ere used as the ex plants in the regenerat ionexperiments.Six factors such as geno types , plant grow th regulators , sonication assisted t reatment and so onw ere studied .The result s show that the high regeneration f requency geno types are Hefeng 35 , Hefeng 25 ,Heinong 40 , the multiple shoot s per explant are 6 .69 , 6 .32 and 5 .98 respect ively .The BA concentrationsrequired in shoot dif ferentiation medium are different among these geno types, in Hefeng 35 , 1 .5mg/l Hefeng 25and in 1 .2mg/l Heinong 40 .But the IBA concentrations in root regenerat ion medium are the same .The fullcotyledo nary is the best explant size for shoo t differentiation .As an antibiotic to restrain the propagatio n ofexcess Ag robacterium , the Cef has no sig nificant inf luence on regeneration .When the t reatment time is shorterthan 12s , the sonicat ion assisted t reatment has no sig nificant influence on the regeneration of shoots .
