Abstract:
Comparative observation on microspore development in cy toplasmic -nuclear male sterile soybean material, such as (N8855 ×N2899)F1 , BC1 male sterile plant s , NJCMS1A and i ts fertile countpart (N2899 ×N8855)F1 , maintainer line , w as made w ith light and transmission electron microscopy .The result show ed theabortion appearance of the dif ferent generatio ns with the same cy toplasm w as similar , pollen abo rtion occurredmajorly f rom late mononucleate stage to binucleate stage .The abno rmal vacuoles in microspo re in (N8855 ×N2899)F1 male sterile anther occurred at late mononucleate microspore stage .Plasmodium shriveled at maturepollen g rain , The partial cy toplasm disinterg rated , and 2 -3 nucleus w ere found in some microspores in BC1sterile plants .Pollen abortion of NJCMS1A occurred at the stage of binucleate pollen .There appeared mo resmall vacuoles in microspo re cy toplasm of NJCMS1A than that of it s maintainer line at the stage that generativecell closed to vegetative cell , starch grains not being produced in cytoplasm .After generative cell w as aw ay f romvegetative cell , part of cytoplasm disordered , generative and vegetative nuclei further degenerated , vacuoles expanded, part of cy toplasm disinteg rated , and the development of endexine w as completely inhibited .Isozymeanaly sis indicated that the fastest cytochrome oxidase active band missed at binucleate stage , w hich w as consistentw ith the result of the pollen abortion phase .Thus , the mechanism of the cytoplasmic -nuclear male sterilityw as probably associated w ith cy tochrome o xidase .
